This research aims to develop a paper based test strip for the detection of SARS-CoV-2 variants from nasal swabs to diagnose SARS-CoV-2 infection and specific variant. It can detect 3 variants simultaneously. Recombinase polymerase amplification is used to multiply the target gene of SARS-CoV-2 variant and Lateral Flow Assay technology is used to detect the amplified targets. In the first phase, researcher reviewed the literature to select the appropriate platform for LFA. Then, the primers for RPA and the probes for LFA were designed. Further, suitable running buffer and components for gold conjugate solution were optimized. Target detection of LFA was proved with synthetic linear templates. Then, researcher will optimize the detection of RPA amplicons in LFA system. Later, analytical sensitivity, specificity and clinical detection with patients samples will be optimized to prove the clinical application of the developed platform.
